Quest 1 - Radamant's Lair
Go to town and find out who's selling what. Along the way, you should meet Atama, who will give you a quest. The sewers have two entrances from town; either one will work:
There are three levels, and Rhadamanthus is at the bottom. Kill him and take the Horadric Scroll, which leads to the next quest.
Quest 2 - Horadric Staff
This quest is very long; you'll complete the third and fourth quests along the way. Take the scroll to Kain, the last of the Horadric line, and he'll translate it. He'll also tell you about the next quest, which consists of three parts: first, you'll need to obtain the Horadric Cube, then the Shaft, and finally the Head of the Staff.
The Horadric Cube is located in the Halls of the Dead. Head out of town to the Rocky Wastes, then to the Dry Hills, where the Halls of the Dead are located. At the entrance, the fight will be to the death, so be prepared to die.
Once you have the Cube in your hands, go see Kain. The Cube has more slots than it needs to hold, so it can be used as a wonderful bag. Experiment and see what it can transform into:
You'll find the spear in the Worm's Lair. This terrifying dungeon is located in the Far Oasis, adjacent to the Dry Hills.
If you prefer a brawl, you'll need a ranged weapon and lightning resistance. The lair's inhabitant will drop a poisonous substance upon death, so avoid standing too close. The spearhead is located in the Temple of the Viper's Claw.
Travel through the Oasis to the Lost City, and from there to the Valley of the Serpents. You'll obtain the spearhead and complete the third quest. Place the found parts (shaft and spearhead) in the cube and press the Transmute button, and you'll receive the completed Horadric Staff.
Quest 3 - Tainted Sun
During your travels (either in the Dry Hills or the Far Oasis), you'll begin the third quest. When everything is plunged into darkness, go to town and speak with Atamoi. She'll tell you what happened. The filthy Talon Snakes seem to not only possess the Arrowhead, but are also the cause of the darkness throughout the land. Thus, having obtained the Arrowhead, you will automatically complete the third quest.
Quest 4 - Arcane Sanctuary
After acquiring the Arrowhead and completing the rest of your tasks, go to town. Kain will tell you what to do and direct you to Jergin, who will then let you into the Palace. Inside the sanctuary, you'll need to find the journal of a dead old man.
To find the sanctuary, go to the palace, go through all three levels of the Harem, then the Palace Cellar, until you find a portal to the Arcane Sanctuary. You'll need to use ranged attacks there, so be prepared.
You'll have to kill the guy guarding the book. He's the Summoner, and this completes the fifth quest. Traditionally, the end of one quest signifies the end of another. Don't forget to read the book!
Quest 5 - The Summoner
The Summoner stands on a platform with fire-spitting ghouls. This is a very bad situation; you'll definitely need a ranged attack. Kill him and read the tome, which will open the gate to the Canyon of the Magi. It will also tell you which of Tal Rasha's tombs you need. Either head straight for it, or search the others for treasure first.
Quest 6 - Tal Rasha's Tomb
This is a very fast level; you're approaching the end of Act II. Find Tal Rasha's tomb. If you accidentally forget which tomb you need, check the quest log.

Proceed through the tomb until you find a passage. There you will also see a place for the Staff. Insert the Horadric Staff in its place, and a wondrous metamorphosis will occur: the final level of the tomb will open. As you pass through the passage, be prepared for a fight. Kill the worm-like fellow—he's quite strong. Explore the surrounding area and talk to Tyrael.
